Edit: It depends on what technologies you know and how much you are willing to invest in learning -- who you are targeting for users. For the chat software projects I've been involved with, I usually wrote the server software in C++ or Java and the front-end clients were Flash, Java, and AJAX.
Flash benefits: Loads quickly, widely deployed, small to download, easy to develop for.
Flash drawbacks: Frequent security issues, Apple doesn't want to support Flash because it allows their users to circumvent their App store, plugin is required for the browser.
Java benefits: Easy to develop for, lots of functionality (full language)
Java drawbacks: Users must have Java installed, JRE is pretty big.
AJAX benefits: Minimal browser requirements (works with mobile devices), easy to develop for
AJAX drawbacks: Various browser issues, polling approach
You also need to consider if your users will be connecting from behind proxy servers (HTTP or transparent proxies) SOCKS proxies don't require anything special on your side.
